Basic COAs have all relevant information on one page. other sample testing analysis information can be found in the samples ELN folders
Full length COAs have multiple pages. Equivalency Values such as "PCBE" & "PCNE". These figures are based on the molecular weights of the two major alkaloids of magic mushrooms & takes the conversion factors into account due to the compounds Phosphorylating or dephosphorylating and both figures as useful to get idea of total potency that maybe felt. "Minors" figure is also given as a total of minor alkaloids to get idea of any entourage effects.
The "Profile" Pie Charts given a visual view of the total compounds found and the ratios between them. This can give idea of entourage effects and complete makeup "profile" of your strain.
The "Bubble Boxes" page as we call it, shows similar figures this time only in milligrams per gram. Its good to have both % percentages and mg/g for the most advised dosing decisions.
The chromatographs give the actual absorbance peaks and retention times based on the method which it was run. These graphs are invaluable to give another visual aid on the absorbance "mAu" score and peaks seen by the HPLC technician. This also can give clues to how the tech ran the sample, what else was picked up and perhaps not reported.
Beneath the graphs are the calibration tables with what compounds were looked for on that method. We have multiple methods and standard methods. We like to make specific method which more efficiently examines certain compounds for more in depth research.
Our PCR/ DNA sequencing amplifies the DNA of your strain making lots of copies. We have spent a lot of time perfecting our SOP for DNA barcoding and have developed a hybrid tech based off Sigrid (YouTube) & Alan Rockefellers' Barcoding techs. Along with our new custom bulk mixed primers, dyes and master mixes we can now do DNA sequencing more efficient & successfully than ever! After amplification we run the DNA via Electrophoresis on a gel stain to make sure the DNA is present and then run Sanger Sequencing of the ITS regions, ITS1, ITS4, LSU, SSU, regions. We then examine and edit any chromatographs and their fastfa files, then Blast the sequence on the GenBank on the Nation Library of medicines website, comparing it to their whole database. If the sequences are matched and align able to others, we can then compare them see how similar and different they are and where they fall onto the phenolic family tree. We then can upload your strain to the GenBank Co-Authored with IntroArts Lab. You can then Blast the sequence, view phenolic trees and other useful related mushroom sub strains. Also use their Genbank Ascension Numbers to link to your labels on products, lab reports, etc. There is indeed value of ITS sequencing of sub strain fungi! More info the better. Potency is NOT everything. Its actually just one thing!
The PsiloQtest by Invented by Domas mentor Dr. Felix Blei of Miraculix Germany. PsiloQtest is a fast reagent kit for determining the alkaloid content easily at home!. This again can give great clues on dosing decisions and knowledge about what genetics you are growing. Results are usually within -/+10% of HPLC data and is a great tool before sending a strain off for full lab work.
We can now use a colorimeter/ spectrometer to determine the scores instead of just using our eyes. In this way we can develop a calibration standard as a community based on this open source device to simple measure the absorbance of the PsiloQtests. This can give us better determination of the 2 major alkaloids separately as it can be measure twice at different stages of the reagent test.
This page shows results of the moisture Analysis & Total contamination load. The moisture content of a biomass affects its potency, sample extraction and degradation factors. This test shows the moisture content before and after drying with regain values and solid mass figures. These are helpful in determining an average and true "Anhydrous scores" of the bio mass. As far as Heavy Metals and pesticide analysis this maybe more relevant to outdoor or wild grows, (particularly cannabis, although maybe indoor to with a lot of use of additives and nutrient booster) It may eventually be valuable however at this time these methods can only be dome with Mass Spectrometry in an ICP MS system not LC system. The ICP MS system used Argon gas which is heated up to a plasma. The compound are then vaporized and detected floating in the plasma. This is equipment we can perhaps get in the future or use another lab for. There maybe some basic PASS/FAIL tests we can do for them and more important for Fungi we believe would be mycotoxins and other mold contaminants which we can test for with Agar /PCR/ Electrophoresis and HPLC.
Calibration curves are a representation of the CRM standards of a given compound at various dilutions plotted on a linear line graph. Your compounds score is often highlighted to see where falls on the graph as compared to the standard. More calibration points are desired for accuracy from lowest to highest range. Some compounds require points to grouped lower or higher than others for precise measuring and calibrations. It is best to fall within the curve points.
In more general terms, a calibration curve is a curve or table for a measuring instrument which measures some parameter indirectly, giving values for the desired quantity as a function of values of sensor output. For example, a calibration curve can be made for a particular analytes concentration.
Many times acronyms are used and people are often confused by their meaning. Where possible we give the full compound name or even the scientific name but many times spacing requires us to use acronyms. Below are some common acronyms used:
